Аннотация: This paper proposes a quantitative model of the reaction-diffusion type to examine the distribution of interferon-alpha (IFN alpha) in a lymph node (LN). The numerical treatment of the model is based on using an original unstructured mesh generation software Ani3D and nonlinear finite volume method for diffusion equations. The study results in suggestion that due to the variations in hydraulic conductivity of various zones of the secondary lymphoid organs the spatial stationary distribution of IFN alpha is essentially heterogeneous across the organs. Highly protected domains such as sinuses, conduits, co-exist with the regions in which where the stationary concentration of IFN alpha is lower by about 100-fold. This is the first study where the spatial distribution of soluble immune factors in secondary lymphoid organs is modelled for a realistic three-dimensional geometry

Аннотация: HIV infection is multi-faceted and a multi-step process. The virus-induced pathogenic mechanisms are manifold and mediated through a range of positive and negative feedback regulations of immune and physiological processes engaged in virus-host interactions. The fundamental questions towards understanding the pathogenesis of HIV infection are now shifting to 'dynamic' categories: (i) why is the HIV-immune response equilibrium finally disrupted? (ii) can one modify the dynamic equilibrium for host benefit? (iii) can one predict the outcome of a system perturbation via antiviral drugs or drugs modulating the host immune response dynamics? Answering these questions requires a major interdisciplinary effort, and in particular, the development of novel mathematical approaches for a coherent quantitative description and prediction of intra-patient HIV evolution, the immunological responses to HIV infection, and the systems level homeostatic regulation of specific effector and regulatory lymphocyte populations in correlation with disease status. Аннотация: Plasmacytoid dendritic cell (pDC)-mediated protection against cytopathic virus infection involves various molecular, cellular, tissue-scale, and organism-scale events. In order to better understand such multiscale interactions, we have implemented a systems immunology approach focusing on the analysis of the structure, dynamics and operating principles of virus-host interactions which constrain the initial spread of the pathogen. Using high-resolution experimental data sets coming from the well-described mouse hepatitis virus (MHV) model, we first calibrated basic modules including MHV infection of its primary target cells, i.e. pDCs and macrophages (Mphis). These basic building blocks were used to generate and validate an integrative mathematical model for in vivo infection dynamics. Parameter estimation for the system indicated that on a per capita basis, one infected pDC secretes sufficient type I IFN to protect 10(3) to 10(4) Mphis from cytopathic viral infection. This extremely high protective capacity of pDCs secures the spleen's capability to function as a 'sink' for the virus produced in peripheral organs such as the liver. Furthermore, our results suggest that the pDC population in spleen ensures a robust protection against virus variants which substantially down-modulate IFN secretion. However, the ability of pDCs to protect against severe disease caused by virus variants exhibiting an enhanced liver tropism and higher replication rates appears to be rather limited. Taken together, this systems immunology analysis suggests that antiviral therapy against cytopathic viruses should primarily limit viral replication within peripheral target organs

Аннотация: Цель. Изучить показатели воспалительного процесса на разных стадиях развития атеросклероза. Материал и методы. У 174 человек в возрасте 21-70 лет выполнено ультразвуковое сканирование брахеоцефальных сосудов на аппарате АЛОКА 5000, для визуализации атеросклеротических бляшек и определения толщины комплекса интима-медиа (КИМ). Методом иммуноферментного анализа (ИФА) в плазме крови определяли количественное содержание С-реактивного белка (CРБ) тест-системой SeroELISA фирмы Diagnostic Systems Laboratories (США), и цитокинов с помощью отечественных тест-систем (ООО Цитокин, СПб): фактор некроза опухолей (ФНОα), интерлейкин-1 (ИЛ-1), ИЛ-4 и ИЛ-8. Результаты. Содержание СРБ и провоспалительных цитокинов ИЛ-1, ИЛ-8, ФНОα зависит от стадии атеросклеротического процесса, степени увеличения толщины КИМ. Увеличение концентрации СРБ до уровня, характеризующего системную воспалительную реакцию, отмечено у лиц с субклиническим течением атеросклероза и у пациентов с постинфарктным кардиосклерозом. Заключение. Существует взаимосвязь между толщиной сосудистой стенки и выраженностью латентного воспалительного процесса, причем качественные изменения в интенсивности воспалительного процесса имеют место на начальных этапах формирования атеросклероза

Аннотация: We investigated the impact of bacterial regulators homoserine lactones (HSLs) and alkylhydroxybenzenes (AHBs) (which are present in human fluids at pico- and nanomolar concentrations) on neutrophile oxidative metabolism. The HSL and AHB effects were determined using a test based on induced luminol-dependent chemoluminescence of neutrophiles in human peripheral blood. In this test, neutrophiles were preincubated with chemical analogs of bacterial autoregulators with different lengths of the hydrocarbon radical, such as HSL · HCl, C6- and C12-HSL, and C1-, C6-, and C12-AHB. We revealed that they suppressed the chemoluminescence and, accordingly, the oxidative metabolism of neutrophiles. This effect was more significant with HSLs than with AHBs. Within each of the two groups, the effect increased with an increase in the length of the hydrocarbon chain of the homologues. High concentrations of long-chain autoregulators of both types produce a cytotoxic effect that is associated with apoptosis in the case of C12-HSL and with cell membrane damage in the case of C12-AHB. The effects of low HSL and AHB concentrations involve their protein-modifying properties and result in changes in the activities of neutrophile oxidative enzymes. To a lesser extent, these effects are due to the pro- and antioxidant activities of HSLs and AHBs, respectively. In light of the results obtained, the HSL and AHB effects are to be considered as a novel mechanism of regulating the activities of cell effectors of natural innate immunity. In symbiotic and parasitic systems, the mechanism involves the bimodal pattern of the effects of HSLs and AHBs that vary depending on their structure and concentrations

The aspiration system of one furnace operates in two regimes and the gases are subjected to single-stage cleaning in a sprayed cyclone-type shaft. Dust removal efficiency is 90%. The suctioned air from the other furnace is cleaned of charge materials in battery cyclones and a sprayed cyclone-type shaft. Cleaning efficiency is 90-93%.

Аннотация: Infections with HIV represent a great challenge for the development of strategies for an effective cure. The spectrum of diseases associated with HIV ranges from opportunistic infections and cancers to systemic physiological disorders like encephalopathy and neurocognitive impairment. A major progress in controlling HIV infection has been achieved by highly active antiretroviral therapy (HAART). However, HAART does neither eliminate the virus reservoirs in form of latently infected cells nor does it completely reconstitute immune reactivity and physiological status. Furthermore, the failure of the STEP vaccine trial and the only marginal efficacies of the RV144 trial together suggest that the causal relationships between the complex sets of viral and immunological processes that contribute to protection or disease pathogenesis are still poorly understood. Here, we provide an up-to-date overview of HIV-host interactions at the cellular, the immune system and the neuroendocrine systems level. Only by integrating this multi-level knowledge one will be able to handle the systems complexity and develop new methodologies of analysis and prediction for a functional restoration of the immune system and the health of the infected host

Аннотация: The analytical system for the determination of the group-A streptococci (SGA) has been constructed. The designed diag-nosticum does not depend on hardware, i.e. it belongs to the group of so-called non-instrumental systems. The suggested pattern in realized based on the original conjugate (diagnostic reagent) that is synthesized on the basis of covalent binding of polyclonal antibodies against group-A streptococci with colloid carbon particles; this conjugate possesses the ability to bind specifically to appropriate ligand. Due to high absorbance of such conjugates in the visible light region it is possible to make direct visual determination of SGA in a sample under study. The reaction is manifested in a form of black dots on a solid phase working surface. The system could be arranged either in the form of direct determination or in a sandwich-test variant. Such analytical systems could be an efficient tool for dispensary, hospital, and pre-clinical testing including extreme clinical and epidemic situations, as well as these could be applied in different kits for home and field diagnostics

Аннотация: Optimal control problems for mathematical models describing HIV infection dynamics in a human body are considered in the paper. An overview of current approaches to solution of control problems for models of HIV dynamics is presented for techniques related to construction of optimal programme (open loop) or positional (feedback) controls for various criteria of control process quality and is based on the Pontryagin's maximum principle and the Bellman's theory of dynamic programming, respectively. In the framework of the theory of positional differential games of Krasovskii, there are constructive and efficient methods of synthesis of controls for different classes of dynamical systems. In this paper we present a formalization of a control problem for HIV infection considered as a corresponding positional differential game. The control algorithm based on the method of extremal shift is applied to the ODEs model of HIV infection. The numerical implementation of the extremal shift method is developed to construct the control taking the system to a neighbourhood of a given trajectory using the information on the dynamics of the guides or leaders.

Аннотация: The variable polyrhythmicity observed in natural, technical, and humanitarian spheres can be the result of mutual desynchronization of conjugate rhythmic processes. One example of desynchronization in medicine is fibrillations of heart ventricles. To bring a patient out of this state, electroshock is applied, which frequently leads to defibrillation and recovery of normal heart rhythm (resynchronization). This phenomenon has analogies in other fields of science. It is known that lunar–solar tides cause subsurface seismic noise. We give an example of detecting such noise in the Ashkhabad seismic survey area. This noise can appear and disappear, and the reason may be certain shock impacts, in particular, earthquakes. Similar results have been obtained in studying lunar seismicity, where tidal phenomena are more strongly pronounced than on Earth.
